Harkness inherited a large fortune from his father, Stephen Vanderburg Harkness, a partner of John D. Rockefeller.

He contributed to the Metropolitan Museum of Art, the New York Public Library, Harvard and Yale universities, and the Columbia-Presbyterian Medical Center in New York City. In 1918 Harkness helped to establish a foundation known as the Commonwealth Fund. The foundation's assets are used mainly for medical research and education, medical and public-health services in rural areas, and mental-health services.
