Elizabeth Holmes Fisher collected Dutch and Flemish masterworks from the 16th through 18th centuries; art from the 19th century American Hudson River School; French Barbizon landscapes; and 17th and 18th century British portraits.
The collection included work by, among others, Jan Breughel, Pieter Codde, Jan de Bray, Marcus Gheeraerts the Younger, Dirk Hals, Claude Lorraine, Gaspar Netscher, Jacob van Ruysdael, Albert Bierstadt, Thomas Cole, Asher B. Durand, Thomas Doughy, Henry Inman, David Johnson, George Inness, Thomas Gainsborough, Sir Peter Lely, Sir Henry R.A. Raeburn, Sir Joshua Reynolds, George Romney, and Benjamin West.
In 1937, Fisher gave a gift to the University of Southern California (USC) to construct and art museum, the Elizabeth Holmes Fisher Art Gallery, now known as the USC Fisher Museum of Art. When founding the museum, she gave the university 74 paintings, drawings, and sculptures. Fisher was the first woman to on the board of USC.
She was married to Walter Harrison Fisher (1866-1926). They had three children: Rachel A. Fisher Yerge Frayram (1887–1971), Ruth L. Fisher Oxnam (1889–1975), and Wayne H Fisher (1892–1962). Her parents were Holmes and Baxter.
